Per oxidas e-act iv ity in m it ogen-stimu - lated lymphocyte culture After incubation of Isopaque-Ficoll separated mononuclear leukocytes in vitro for 3 days, the cell suspension was depleted of peroxidase activity, whether they were cultured with mitogens or not. The effect of nylon fibre column fractionation on the peroxidase posi- tive cells In the nylon fibre fractionation experiments four different blood donors were tested. Before fractionation of the Isopaque/Ficoll separated cells, the peroxidase positive cells were on average 19% (range 197o-29/o). (Fig. 1). In contrast, cell suspensions fractionated on a nylon column contained practically no peroxidase posi- tive cells (< 0. 9$) nor cells with monocytic morphology as determined by differential count- ing of the cell suspensions stained with May- Gríinwalds Giemsa. After fractionations the percentage of EA-RFC was slightly reduced indi- cating that a small proportion of the peroxidase positive cells might also form EA-rosettes. This was supported by peroxidase-staining of the EA- rosette preparations made before nylon fibre fractionation which showed that on an average. 23.6% of the EA-rosette forming cells were peroxidase positive (range 21%-28/o). After nylon fibre fractionation no EA-rosettes were peroxidase positive (Fig. 2a) and the cytotoxic activity of the cell suspensions was actually higher, in all four experiments (mean CI 69.0, and range 54.1-80.4) than before fractionation. (Mean CI 39.4 and range 22.5-69.3) (Fig. 2b).
