294 LÆKNABLAÐIÐ untranslated region, in the coding sequence, and in the 3’ flank did not significantly affect PNP expression. Two fortuitous splice sites in the PNP complementary strand co-localized with the enhancer in intron-1. Shortening of the 2.9 kb intron-1 to 855 bp had no effect on PNP expression. Further shortening of intron-1 to 338 bp by eliminating both the enhancer and the fortuitous splice sites abolished PNP expression. Expression was partially restored if the enhancer was reinserted in the reverse orientation upstream of the PNP promoter or in the 338 bp intron-1. Elimination of several sequences associated with deletions during retroviral-mediated gene transfer thus provided PNP genes which retained function and may be transduced intact once tested in a retroviral vector.
