með óreyndum athuganda og greiningartækjum er
gefa lélega mynd. Reyndur athugandi getur hins vegar
nálgast rétta greiningu með nokkurri nákvæmni þrátt
fyrir miður góð myndgæði.
1) Hér er gert grein fyrir framgangi sýkingar í óbólu-
settu rottunum, rotturnar sem voru bólusettar , þjóna
ekki hlutverki við stöðlun á dýramódelinu og verður
gert frekari skil í erindi um “Ónæmisfræði miðeyrna-
sýkinga í rottum”.
Lipoxygenasa-ferlar og stjórn frumufjölgunar

Inngangur: Lipoxygenasar (LO) mynda eicosanoiða
úr arachidonic sýru og finnast þrjú ísóensím, 5-, 12-
og 15-LO sem mynda HETE og leukotríen. 5-LO örv-
ar vöxt ýmissa æxlisfruma en 12-LO auðveldar ífar-
andi vöxt. Sýnt hefur verið fram á virkjun MAP
kinasans ERKT/2 með HETE. Protolichesterinic sýra
(PA) úr fjallagrösum hindrar 5-LO en fyrri vinna okk-
ar sýndi vaxtarhindrandi áhrif PA á 12 illkynja æxlis-
frumulínur. Áhrif PA á adenocarcinoma-línur úr ristli
benda til að hún hindri einnig 12-LO. Markmið þess-
arar rannsóknar var að kanna hvort PA miðli vaxtar-
hindrandi áhrifum sínum í gegnum MAP kinasa feril-
inn og að kanna hvort 5- og 12-LO séu tjáð í þremur
illkynja æxlisfrumulínum sem PA hindrar.
Efniviður og aðferðir: T47D úr brjóstakrabbameini,
PANC-1 úr briskrabbameini og WIDR úr ristilkrabba-
meini fengust hjá ATCC. PA var einangruð við lyfja-
fræðideild Háskóla íslands. Frumurækt fyrir MAP
kinasa tilraunir var svelt og örvuð með sermi, með og
án PA. Prófað var fyrir fosfóruðum ERKl/2 með
immunoblotti. Tjáning á 5- og 12-LO var metin með
mótefnalitun.
Niðurstöður: MAP kinasar voru misvirkir eftir
frumulínum og stöðu í frumuhring. PA hindraði MAP
kinasa örvun í PANC-1. Mótefnalitanir sýndu að 12-
LO var í umfrymi en 5-LO í kjarnahimnu, sterkast í
PANC-1 en veikast í WIDR. 5-LO var einnig í kjarna
hjá T47D og var það breytilegt eftir staðsetningu í
frumuhring.
Ályktun: PA miðlar líklega vaxtarhindrandi áhrifum
sínum í gegnum MAP kinasa ferilinn í PANC-1. Við
virkjun færist 5-LO að kjarnahimnunni og því er 5-LO
líklega á virku formi í PANC-1 og T47D en minna 1
WIDR. Styrkleiki litunar er í beinu samhengi við
næmi frumulínanna fyrir PA.
Effects of mifochondrial gradient
disruptors and maintainers on Leydig cell
steroidogenesis.

Introduction: Mitochondria have an electrochemical
gradient (((m) across their inner membrane as a result
of the activity of thc electron transport chain. This
gradient is necessary for many mitochondrial funct-
ions, including steroidogenesis and expression of the
Steroidogenic Acute Regulatory protein (StAR). The
objective of tliis study was to examine how substances
that disrupt ((m affect steroidogenesis and the levels
of StAR. Also, it was interesting to see if substances
that are thought to protect the electrochemical gradi-
ent could maintain steroidogenesis and StAR levels in
the presence of mitochondrial disruptors.
IVIaterials and methods: Mouse tumor Leydig cells
(MA-10) were treated with 8-Br-cAMP for 3 h +/-
agents tliat target and disrupt mitochondria. These
agents included CCCP, nigericin, oligomycin, Na+
arsenate and H202. Three groups of MA-10 cells
were also exposed to H202 only; a control group, a
group that had been treated with Cyclosporin A and a
group that had been transfected with the Bcl-2 gene,
both of which are supposed to keep the ((m intact.
Post treatment, media were collected and subjected to
progesterone radioimmunoassay (RIA) and levels of
StAR protein determined by western blot analysis.
Results: RIA data suggested that progesterone prod-
uction was significantly decreased after treatment
with all nritochondrial gradient disruptors tested.
Western blot analysis of StAR protein showed that all
disruptors either decreased one or both forms of StAR
or totally inhibited its expression. Results from the
Cyclosporin A and Bcl-2 experiments are not avail-
able at this point.
Discussion: These results demonstrate that perturbing
mitochondria by disrupting ((m will reduce ex-
pression and normal processing of StAR and prevent
steroidogenesis.
